Function/Duties of Position

A funded postdoctoral position is available at Oregon Health & Science University (OHSU) in Portland, Oregon. The laboratory studies therapeutic vulnerabilities in oncogene-driven cancers using proteomic, cellular, and translational approaches. The position is supported by a funded research program investigating how targeted kinase inhibitors remodel the tumor cell surface proteome and create new opportunities for antibody-drug conjugate (ADC)-based therapies in ROS1 fusion-positive cancers. The postdoctoral scholar will lead studies utilizing quantitative surface proteomics, flow cytometry, and functional drug-response assays to identify novel therapeutic targets and mechanisms of response to ADCs.

 

Quantitative Proteomic Analysis of the Tumor Surfaceome
Lead studies characterizing cell-surface protein remodeling in response to targeted kinase inhibitors. Perform cell-surface biotinylation, sample preparation for LC-MS/MS analysis, and downstream analysis of quantitative proteomic datasets.A nalyze surfaceome datasets to prioritize therapeutic targets.

 

Functional Validation of Candidate Surface Targets
Validate prioritized cell-surface proteins using flow cytometry, immunoblotting, immunofluorescence microscopy, and complementary molecular approaches. Quantify antigen abundance and assess changes induced by targeted therapies.

 

Therapeutic Response and Cell Viability
Studies Design and execute experiments evaluating kinase inhibitors, antibody-drug conjugates, and combination therapies. Perform cell viability, proliferation, apoptosis, and cell-cycle assays and analyze dose-response and synergy relationships.

 

Data Analysis and Interpretation: Analyze and integrate proteomic, flow cytometric, imaging, and pharmacologic datasets. Perform statistical analyses, prepare publication-quality figures, and contribute to manuscripts and grant applications.

 

Scientific Communication: Prepare and present research findings at scientific meetings and in peer-reviewed publications. Draft manuscripts, contribute to grant preparation, and sustain clear written and verbal communication for internal and external audiences.

Required Qualifications

  • PhD or equivalent doctoral degree, awarded or expected before the start date.
  • 0 to 5 years of postdoctoral experience.
  • Demonstrated, hands-on proficiency in mammalian cell culture, including sterile technique and long-term maintenance of established and patient-derived cancer cell lines.
  • Demonstrated proficiency across core molecular biology and biochemistry, including molecular cloning and site-directed mutagenesis, stable-line generation, quantitative immunoblotting, and immunoprecipitation.
  • Ability to independently design, execute, troubleshoot, and interpret well-controlled experiments, and to analyze and integrate multi-modal datasets with appropriate statistical treatment, with meticulous record-keeping and attention to rigor.
  • Evidence of scientific productivity and follow-through, demonstrated by at least one first-author or co-first-author peer-reviewed publication or preprint.
  • Resourcefulness, creativity, and independent problem-solving, including the ability to diagnose failed experiments, locate solutions, and adapt approaches independently.

Preferred Qualifications

  • Prior research in cell signaling, biochemistry, cancer biology, or a related mechanistic discipline.
  • Experience with mass spectrometry-based proteomics, quantitative proteomics, phosphoproteomics, or cell-surface proteomics. 
  • Experience preparing samples for LC-MS/MS analysis and interpreting proteomic datasets. 
  • Expertise in flow cytometry and quantitative immunophenotyping. 
  • Experience with cell viability, proliferation, apoptosis, and drug-response assays. 
  • Familiarity with antibody-drug conjugates, targeted therapeutics, and cancer biology. 
  • Experience with patient-derived cancer models. 
  • Working knowledge of proteomic analysis software such as MaxQuant, Perseus, Skyline, or equivalent.
